DMEM (Dulbecco’s Modified Eagle Medium) یک محیط پایه است که به طور گسترده ای در کشت سلول مورد استفاده قرار می گیرد این محیط، از رشد بسیاری از سلول های مختلف پستانداران پشتیبانی می کند،مناسب ترین محیط برای بررسی فنوتیپ سلول های چسبندهَ همانند فیبروبلاست های اولیه، سلول های عصبی، سلول های گلیال ،سلول های عضلانی صاف و بسیاری از این قبیل به خوبی در این محیط قابل رشد هستند.

این پودر پس از محلول شدن پروتئین، لیپید یا فاکتورهای رشد ندارد،بنابراین نیاز است مکمل‌های رشد را به آن اضافه کنیم. می‌توان این محیط را معمولاً با 10٪ سرم جنین گاوی (FBS) غنی‌سازی و سپس بی‌کربنات سدیم به میزان 3.7 گرم در لیتر به‌عنوان بافر استفاده کرد،همچنین برای حفظ PH نیاز است تا 5–10% CO₂ به محیط اضافه شود.

DMEM is unique from other media as it contains 4 times the concentration of amino acids and vitamins than the original Eagle’s Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate. Using DMEM
DMEM contains no proteins, lipids, or growth factors. Therefore, DMEM requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). DMEM uses a sodium bicarbonate buffer system (3.7 g/L) and therefore requires a 5-10% CO₂ environment to maintain physiological pH. Powder forms of Gibco™ cell culture medium require sodium bicarbonate supplementation, pH adjustment, and filtration at the time of preparation (see protocol for details)
